rAAV vector production core — IBCCF/UFRJ
Vector Core
The first national recombinant adeno-associated virus (rAAV) vector production core was established by Prof. Hilda Petrs-Silva at IBCCF, enabling preclinical and clinical gene therapy research in Brazil without dependence on imported vectors. The core combines large-scale technical capacity with expertise in capsid engineering and in vivo biological validation.

Custom-built vectors
Available serotypes
AAV1, AAV2, AAV5, AAV8, AAV9, and tyrosine-mutants (Y444F, Y500F, Y730F).
Vector construction
Design and cloning of rAAV-ITR plasmids with specific promoters (CBA, CMV, custom mini-promoters for ganglion cells).
Large-scale production
Triple transfection in HEK293 with iodixanol gradient purification and qPCR titration.
In vivo validation
Transduction analysis in retina and CNS via intravitreal, intravenous, or intrastriatal injection — with GFP/luciferase readout.
Advanced engineering
Capsids with targeting peptides (phage display), CRISPR-Cas9, and CRISPR-a packaged in rAAV.
